Research summary
Nicotinamide adenine dinucleotide (NAD⁺) is a vital metabolite that plays a central role in a broad spectrum of physiological processes, including energy metabolism, DNA repair, and cell survival. Dysregulation of NAD⁺ homeostasis has been increasingly recognized as a hallmark of aging and a contributing factor in numerous pathological conditions, such as cancer and neurodegenerative disorders. The recent identification of the mitochondrial NAD⁺ transporter SLC25A51 has catalyzed growing interest in the spatial dynamics of NAD⁺ metabolism, particularly the functional significance of its subcellular compartmentalization. The primary objective of my current research is to elucidate how mitochondrial NAD⁺ transport supports neuronal resilience under various stress conditions. Using a combination of pharmacological modulation and genetic engineering approaches, I am investigating whether the overexpression of distinct mitochondrial NAD⁺ transporters can confer neuroprotection against insults such as DNA damage, mitochondrial dysfunction, and NMDA receptor-mediated excitotoxicity. This work aims to advance our understanding of NAD⁺-dependent mechanisms of neuroprotection. We are hopeful that it will ultimately inform the development of novel therapeutic strategies for brain injury and neurodegenerative diseases and promote healthy aging.
